The oncogenic tyrosine kinase (TK) and ras-like GTPase products of the epidermal growth factor receptor (EGFR) and the c-Ki-ras proto-oncogene are key proteins in the regulation of normal cell growth and differentiation. Signals from the oncogenic TK and ras, either independently or in concert, have been shown to transform cells in vitro. Transfection studies in which c-Ki-ras cDNA was co-transfected with cDNA for the EGFR TK, or vice versa, in cells expressing normal levels of each protein, suggested that the two proteins acted in a cooperative fashion to stimulate cell proliferation. To explore this further, we examined the effects of co-expression of c-Ki-ras and TK on the phosphatidylinositol 3-kinase (PI 3-kinase) and ras-dependent signal transduction pathways in Rat-1 cells. We found that the c-Ki-ras protein increased the levels of phosphorylated proteins in the PI 3-kinase and ras pathways in a manner similar to that for the EGFR TK. We propose
